چکیده

In terms of type I error rate, Log rank test and Wald test give similar results. We can see that when the randomization ratio is 1:1 for an event size of 600, no inflation of types I error rate is observed. However when the randomization ratio is 1:2 for an event size of 600, the type I error rate is inflated to about 0.027. The inflation becomes smaller when the sample size increases. However the inflation is not observable only when the sample size is impractically big. When the randomization is further imbalanced to 1:3 for sample size of 600, the type I error is further inflated to about 0.028. Introduction

Sequential boundaries approach in clinical trials with unequal allocation ratios

BACKGROUND In clinical trials, both unequal randomization design and sequential analyses have ethical and economic advantages. In the single-stage-design (SSD), however, if the sample size is not adjusted based on unequal randomization, the power of the trial will decrease, whereas with sequential analysis the power will always remain constant. Our aim was to compare sequential boundaries appro...

Randomization in cancer clinical trials: permutation test and development of a computer program.

When analyzing cancer clinical trial data where the treatment allocation is done using dynamic balancing methods such as the minimization method for balancing the distribution of important prognostic factors in each arm, conservativeness occurs if such a randomization scheme is ignored and a simple unstratified analysis is carried out. In this paper, the above conservativeness is demonstrated b...

QSAR Modeling of COX-2 Inhibitory Activity of Some Dihydropyridine and Hydroquinoline Derivatives Using Multiple Linear Regression (MLR) Method

COX-2 inhibitory activities of some 1,4-dihydropyridine and 5-oxo-1,4,5,6,7,8-hexahydroquinoline derivatives were modeled by quantitative structure–activity relationship (QSAR) using stepwise-multiple linear regression (SW-MLR) method. The built model was robust and predictive with correlation coefficient (R2) of 0.972 and 0.531 for training and test groups, respectively. The quality of the mod...

Blocked Randomization with Randomly Selected Block Sizes

When planning a randomized clinical trial, careful consideration must be given to how participants are selected for various arms of a study. Selection and accidental bias may occur when participants are not assigned to study groups with equal probability. A simple random allocation scheme is a process by which each participant has equal likelihood of being assigned to treatment versus referent ...
